Among people diagnosed with Alzheimer’s, researchers recognize familial forms of the disease and sporadic cases. This shifting appreciation of inherited risk, researchers say, is due to a better understanding of the role of a fourth gene that carries the blueprints to make a lipid-carrying protein called apolipoprotein E, known as APOE. One known as APOE2 is thought to be protective against the development of Alzheimer’s disease. They also compared people with two copies of APOE4 to people with other inherited forms of the disease — early-onset autosomal dominant Alzheimer’s disease (ADAD) and Down syndrome-associated Alzheimer’s disease (DSAD). Gene testing isn’t currently recommendedIt is also likely to change how people who carry the APOE4 gene are diagnosed and treated.

DES MOINES, Iowa (AP) — Three senior members of a super PAC backing Florida Gov. Ron DeSantis left the group on Saturday, the latest sign of instability within the 2024 hopeful's political operation just six weeks before the Iowa Republican caucuses. Longtime DeSantis ally Scott Wagner, who had been a member of the group's board, was named interim CEO and board chairman. DeSantis' campaign has relied heavily on Never Back Down for basic campaign functions, though the two sides cannot directly coordinate under federal campaign finance rules. Never Back Down has more staff working in Iowa, more than two dozen, than any campaign or super PAC and have been organizing ahead of the precinct-level caucuses since June.

His allies have questioned the messaging and depth of the ad-buying by the super PAC, according to two people briefed on the matter. A third said some allies believed Mr. DeSantis was being blamed by voters for the negative spots. Since they came into existence nearly 15 years ago, super PACs have traditionally handled negative messaging and advertising against a candidate’s rival. The group’s main strategist, Jeff Roe, and a board member, Scott Wagner, who is a college friend of Mr. DeSantis, had a heated argument during a discussion about money, according to people briefed on the matter.

Washington, DC CNN —Americans are taking notice of a possible government shutdown and the uncertainty of ongoing labor strikes, according to the University of Michigan’s latest consumer sentiment survey. The university’s Consumer Sentiment Index edged lower by 1.4 points in September, according to a final reading. That would mean some federal workers become furloughed and agencies operate with skeleton crews, hamstringing agencies’ ability to provide crucial government services. The ongoing United Auto Workers strike is also not expected to end soon. With so much turmoil, Americans will likely feel uneasy in the coming weeks, which could affect their spending.
